摘要
研究了不同激素处理、不同花粉发育时期、不同低温预处理和高温预培养对丹参花药愈伤组织诱导的影响。结果表明:最佳的培养基激素组合为MS+2.0 mg/L6-BA+0.2 mg/L NAA;单核靠边期的花药愈伤组织诱导率最高;4℃低温预处理2 d最有助于愈伤组织发生;高温预培养不利于愈伤组织诱导率的提高。
Several factors such as different hormone treatments,pollen developmental stages,low-temperature pretreatment and high-temperature pre-incubation were studied on anther callus induction of Salvia miltiorrhiza.The results showed that 2.0 mg/L 6-BA + 0.2 mg/L NAA was the best hormone combination;the highest anther callus inductivity appeared at the uninucleate eccentric stage;the pretreatment at 4℃ for 2 days was the most conductive to callus occurrence,while pre-incubation at 35℃ was not conductive to the increase of callus induction rate.
