摘要
作者采用PCR和DNA测序方法,研究了齿爪金龟属三亚属核糖体DNA(rDNA)的第一内转录间隔区(ITS1)基因序列的特征,并以索鳃金龟属(Sophrops Fairmaire)做外群,构建了分子系统树。研究结果表明:7种金龟甲ITS-1片段序列中,T,A,G,C的含量分别为20.3%、22.9%、29.4%、27.4%,A+T的含量明显低于G+C的含量。分子系统树显示,三亚属的系统发育关系为(齿爪金龟亚属+狭肋金龟亚属)+脊头金龟亚属,齿爪金龟属三亚属间的遗传距离要远远小于与索鳃金龟属间的距离。
The sequences character of nuclear ribosome gene ITS-1 on the subgenera of Holotrichia Hope was studied by PCR and DNA sequence-deteimination.As the outgroupe of Sophrops sp.,molecular phylogenetic trees were established.The average contents of T,A,G,C were 20.3%,22.9%,29.4%,and 27.4%,respectively.The content of A+T was significantly lower than that of C+G.The trees showed that the relations of the phyligentic were(Holotrichia.Holotrichia Reitler+H.Eotrichia Medvedev)+H.Pledina Reitle.The distances among the three subgenera of Holotrichia Hope was significantly shorter than that between anyone of the three and the Sophrops sp..
