摘要
目的探讨肿瘤坏死因子α(TNF-α)与围绝经期的关系,研究中药补肾复方对围绝经期大鼠卵巢TNF-α的影响。方法根据重量和阴道细胞角化指数将大鼠分为7组,每组10只,对照组服用六味地黄汤,模型组、青年组与老年组每日灌服4ml的生理盐水,其余3个组分别服用补肾复方高、中、低剂量,4周后取卵巢标本,采用SABC法检测大鼠卵巢TNF-α积分光密度。结果 TNF-α阳性目标积分光密度值:与青年组比较,除补肾复方高剂量组无统计学差异(P>0.05)外,其余各组均有统计学差异(P<0.05);与模型组比较,各组均有统计学差异(P<0.05),用药各组显著降低,补肾复方各组随着剂量的增加而降低;与对照组比较,补肾复方高剂量组降低显著,有统计学差异(P<0.05);老年组最高,与其他各组有统计学差异(P<0.05)。结论卵巢TNF-α水平随年龄增长而增加,可能是卵巢衰老的一个促进因子,补肾复方使围绝经期大鼠卵巢功能增强,抑制了TNF-α水平,延缓了卵巢的衰老。
Objective To investigate the effects of Chinese herbal medicine on the expression of TNF-α(tumor necrosis factor-α)in rat ovaries during perimenopause period and the relationship between TNF-α and perimenopausal period.Methods Female Wistar rats were randomly divided into 7 groups,according to weight and cardiac index of vagina cell,and each group has 10 rats.Control group were given Rehmanniae Decoction of Six Ingredients,model group,young group and older age-group were given 4ml normal saline,others were given invigorating the kidney compound recipe.The ovaries of rats was used to detect the integral photodensity of TNF-α by SABC(Strept Actividin-Biotin Complex)after 4 weeks.Results Compared with young group,except the group of high dose invigorating the kidney compound recipe(P〉0.05),others have significant deviation(P〈0.05);compared with model group,each group has significant deviation(P〈0.05),the expression of medicine groups are all degrade,the masculine target integral photodensity of invigorating the kidney compound recipe groups are degrade with the dose increased;compared with control group,the expression of high dose invigorating the kidney compound recipe group is significant degrade,the result has statistical significance(P 0.05),the integral photodensity of older age-group is very high,and has significant deviation(P〈0.05).Conclusions Invigorating the kidney compound recipe enhances rat ovaries function during perimenopause priod,restrains the level of TNF-α,delay the apolexis of ovaries.
