血小板采集后不同时间冷冻保存对其质量的影响 被引量:5

摘要 目的:观察血小板采集后不同时间冷冻保存对其质量的影响。方法:血小板采集后6h,24h,72h和96h加5%的DMSO于-80℃超低温冰箱冷冻保存。3个月后取出解冻,全自动生化细胞分析仪检测血小板计数(Plt)、血小板平均体积(MPV)。乳酸脱氢酶(LDH)试剂盒检测血小板中LDH的含量,流式细胞仪检测血小板膜糖蛋白CD62p的表达。结果:血小板采集后6h和24h冷冻,Plt、MPV、pH、LDH和CD62p表达与新鲜血小板无明显差异,而采集后72h和96h冷冻血小板Plt、MPV和LDH含量明显降低,而pH和CD62p表达较新鲜血小板明显升高。结论:血小板采集后长时间放置后冻存会导致血小板激活,采集后应尽快冷冻保存,以保证冰冻血小板的质量。 Objetive: To study the quality of cryopreserved platelet after being collected in different time point. Methods: The platelets were preserved in -80℃ ultra cold freezer with 5% DMSO after being collected in 6, 24, 72 and 96 h. The platelets were thawed after three months. The blood platelets count (Pit) and mean platelet volume (MPV) were measured by automatic biochemical analyzer. Lactate dehydrogenase (LDH) was assayed by LDH kit and platelet glueoprotein CD62p was detected by flow cytometry. Results: The Plt、MPV,pH,LDH and CD62p of blood platelet showed no differences cryopreserved 6 h and 24 h after platelet collection compared with fresh blood platelet. However, the blood platelet cryopre- served after 72 h and 96 h showed significantly increase in the biomarkers of Plt, MPV and LDH, and decrease of pH and the expression of CD62p compared with fresh blood platelet. Conclusion: The platelet can be activated after collection for a long time. In order to guarantee the blood platelet quality, the platelet should be eryopreserved as soon as possible after collection.
