摘要
目的:研究败酱科败酱属糙叶败酱Patrinia scabra Bunge.的根与根茎的化学成分,探讨其抗菌、抗肿瘤物质基础。方法:采用柱色谱以及重结晶等方法进行分离纯化,根据理化性质及多种波谱方法鉴定化合物结构,并对部分化合物进行琼脂稀释法抑菌试验和MTT法细胞毒性测试。结果:鉴定了12个化合物,分别为:1,3,6,8-四羟基蒽醌(1),lunatin(2),1,3,6-三羟基-2-甲基蒽醌(3),乌苏酸(4),(+)-nortrachelogenin(5),patriscabrol(6),vibutinal(7),11-ethoxyviburtinal(8),Xanthopurpurin(9),1,3,6-三羟基-2-甲基蒽醌-3-O-(O-6’-乙酰基)-新橙皮糖苷(10),1,3,6-三羟基-2-甲基蒽醌-3-O-新橙皮糖苷(11),山柰酚-7-O-α-L-吡喃鼠李糖苷(12)。化合物5、6、10对肝癌细胞HepG2增殖抑制作用的IC50分别为0.116mol.L^-1、7.65×10^-4mol.L^-1、4.07×10^-3mol.L^-1,对肺腺癌细胞A549增殖抑制作用的IC50分别为2.13mol.L^-1、3.17×10^-4mol.L^-1、1.15×10-3mol.L^-1。化合物4对链球菌的MIC为2.5μmol.mL-1,化合物5对变形菌、沙门菌MIC分别为7.5、5μmol.mL-1,化合物6对变形菌、链球菌、沙门菌的MIC分别为5、2.5、1.25μmol.mL-1。结论:化合物8为首次从糙叶败酱分得,化合物1-3、5、7、9-12为首次从该属植物中分得,其中蒽醌类化合物为首次从败酱属中发现。化合物6具有一定的细胞毒性和抗菌活性。
Objective:To study the chemical constituents of the root and rhizome of Patrinia scabra Bunge.,and investigate the anti-bacterial or anti-tumor active ingredients.Method:The chemical constituents were isolated and purified by various column chromatographic and recrystallization methods.The structures were identified by chemical and spectroscopic methods.Some compounds’ anti-bacterial activity were tested using agar dilution method and cytotoxicity by MTT method.Result:Twelve compounds were isolated and identified as follows:1,3,6,8–tetrahydroxy-9,10-anthraquinone(1),lunatin(2),1,3,6-trihydroxy-2-methyl-9,10-anthraquinone(3),ursolic acid(4),(-)-nortrachelogenin(5),patriscabrol(6),vibutinal(7),11-ethoxyviburtinal(8),xanthopurpurin(9),1,3,6-trihydroxy-2-methyl-9,10-anthraquinone-3-O-(6’-O-acety1)-α-L-rhamnosyl-(1-2)-β-D-glucoside(10),1,3,6-trihydroxy-2-methyl-9,10-anthraquinone-3-O-α-rhamnosyl-(1-2)-β–D-glucoside(11),campherol-7-O-α-L-rhamnoside(12).The IC50 to HepG2 cell of compound 5,6 and 10 were 1.16×10^-1 mol.L^-1,7.65×10^-4 mol.L^-1,4.07×10^-3 mol.L3 and the IC50 to A549 were 2.13 mol.L^-1,3.17×10-4 mol.L-1,1.15×10^-3 mol.L-1.MIC The of compound 4 to Streptoccus hemolyticus was 2.5 μmol.mL-1,MIC of compound 5 to Proteus vulgaris,Salmonella typhi,Escherichia coli were 7.5,5,5 μmol.mL-1 and MIC of compound 6 to Proteus vulgaris,Streptoccus hemolyticus,Salmonella Typhi and Escherichia coli were 5,2.5,1.25 and 10 μmol.mL-1,respectively.Conclusion:Compound 8 was isolated from the plant for the first time.Compounds 1-3,5,7,9-12 were isolated firstly from the genus of Patrinia.It’s the first time to report the presence of anthraquinones in the Patrinia genus.Compound 6 had certain cytotoxicity to HepG2 cell and anti-bacterial activity.
