9个牡丹品种叶片、叶柄中总酚含量及多酚氧化酶活性的测定 被引量:3

摘要 对9个牡丹品种的叶片外植体进行水冲洗处理,测定了其总酚含量和PPO活性。结果表明:牡丹不同品种间的总酚含量存在显著差异,叶柄的总酚含量比叶片低,经水冲洗1.0h后总酚含量最低;不同品种间PPO活性无显著差异,经水冲洗0.5、1.0、2.0、6.0h后,牡丹外植体的PPO活性比未冲洗的对照明显降低;外植体材料经水冲洗处理0.5~1.0h时PPO活性最低,此时材料适合进行组织培养。 Explants propagated from 9 species was used.The total phenol content and the activity of PPO explants were determined.The results showed that the total phenol content was different in different cultivars and in different tissues.The PPO actibity was reduced obviously after 1 h water washing.The total phenol content and PPO activity were lower than natural pretreatment.They were reduced obviously after 0.5,1.0,2.0 and 6.0 h water washing.The PPO activity of the tissue culture was the lowest after 0.5~1.0 h water washing,and the material was suitable for tissue culture.
