摘要
通过检测免疫抑制的F1小鼠和杂合子F2裸鼠对Friend小鼠白血病病毒(Fr.MuLV)感染的敏感性,探讨免疫状态对Fv-4基因(特别是对Fv-4基因杂合子)抗FriendMuLV作用的影响。经腹腔接种Fr.MuLV病毒液攻击小鼠或F2裸鼠后,检查其脾脏中Fr.MuLV的增殖及其发病情况。结果表明,免疫抑制的F1小鼠和杂合子F1裸鼠都可被Fr.MuLV感染,脾脏中有病毒增殖;并且,约2/3的杂合子F2裸鼠与BALB/C小鼠一样,在接种病毒后1个月内发病或死亡。说明机体的免疫状态可影响Fv-4基因,特别是Fv-4基因杂合子的抗Fr.MuLV作用。提示在进行基因治疗时要考虑机体的免疫状态等因素。
We have been detected the susceptibility of immune suppressive F1 mice and F2 nude mice carrying Fv 4 gene heterozygous to infection of Friend murine leukemia virus(Fr.MuLV),for investigate the effect of immune states on the Fv 4 antiviral gene resistant to Fr.MuLV infection in this paper.The F1 mice were the offspring of Fv 4 mice (Fv 4 gene homozygous)and BALB/C mice,gene type is shown as Fv 4 r/- .F2 nude mice were expected that by mating between brother and sister of F1 hybrid nude mice,which was derived that by mating Fv 4 mice and BALB/C nude mice.Fr.MuLV was injected intra abdominal to the F1 mice which had been given immune inhibitor FK506 and F2 nude mice. Result showed that Fr.MuLV could infect F2 nude mice carrying Fv 4 gene heterozygous and F1 mice which had treated with immune inhibitor FK506,and Fr.MuLV could propagated in mice spleen,and caused leukemia in the 2/3 of F2 nude mice carrying Fv 4 gene heterozygous.This result suggested the sensitivity of Fv 4 gene heterozygous mice on infection of Fr.MuLV was determined by murine ecological elements such as immune states.The antiviral effect of Fv 4 antiviral gene waw influenced by immune states of mouse.
