摘要
目的探讨前列腺基质细胞对肿瘤发生的作用。方法将BALB/c裸鼠分为两组,每组10只,对照组(D组)裸鼠皮下单纯注射前列腺癌细胞株DU145细胞,实验组(D+P组)混合注射DU145细胞和前列腺外周带分离的基质细胞(PrSC),注射后第9周收集肿瘤组织,记录肿瘤重量。采用TUNEL法检测凋亡细胞的数量,采用免疫组织化学法标记α-SMA、Ki67和P53阳性细胞,分别检测肿瘤组织中基质细胞和血管数目,增殖细胞的数目和P53蛋白的表达。结果D+P组瘤重、血管数量和增殖细胞多于D组,但凋亡细胞和P53的表达两组之间差异并无统计学意义。结论基质细胞在肿瘤产生中发挥了重要作用,PrSC的作用可能是促进肿瘤细胞增殖和血管生成,对细胞的凋亡影响较小。
Objective To investigate the role of prostate stromal cells in the tumor progression. Methods Twenty BALB/c nude mice were randomized divided into two groups. The mice in control group (D group) were subcutaneously injected with prostate cancer cell line DU145. While the mice in experimental group (D+P group) were injected with a mixture of DU145 cells and prostate stromal cells (PrSC) derived from prostate peripheral zone. Nine weeks later, the mice were sacrificed and the tumor tissues were collected and weighted. TUNEL stain was used to detect the apoptotic cells. α-SMA, Ki67 and P53 were detect by immunohistochemical method to evaluate vascular, proliferating cells and the expression of P53, respectively. Results The weight, angiogenesis and cell proliferation of tumor in D+P group were all higher than those in D group. But there was no difference in apoptotosis and P53 levels between the two groups. Conclusions The stromal cells might play an important role in the tumor progression. PrSC might promote tumor cell proliferation and angiogenesis, but has less effect on apoptosis.
