摘要
目的探讨人前列腺癌组织和细胞株中环氧化酶的表达情况,研究环氧化酶抑制剂对丁肿瘤细胞生长的影响和作用机制。方法用Trizol提取正常前列腺组织,前列腺增生组织和前列腺癌细胞株PC-3、LNCaP中总RNA,用半定量RT—PCR法分别检测各组织细胞中cox-1、cox-2基因的表达水平。用不同配制浓度的环氧化酶抑制剂对人前列腺癌细胞株PC-3和LNCaP的培养进行干预,观察细胞的存活率,并用Western blot分析细胞中COX-2蛋白的表达。结果cox-1基因在正常前列腺组织、前列腺增生组织和前列腺癌组织中均有表达。cox-2基因在正常前列腺组织中未见表达,在前列腺增生组织中表达较弱,在前列腺癌组织中有明显表达。cox-1基因和cox-2基因在PC-3和LNCaP细胞株中都有高表达。当在不同时间段使用不同浓度的环氧化酶抑制剂干预以后,细胞中COX-2蛋白表达出现下调,前列腺癌细胞的存活率有下降,这种下降与干预时间和浓度有相关性。结论人前列腺癌细胞中存在着环氧化酶的高表达。环氧化酶抑制剂能影响前列腺癌细胞的生长。可以考虑将环氧化酶抑制剂用于治疗前列腺癌。
Objective To determine the expression of cyclooxygenase in prostate carcinoma cells and to investigate the mechanism of the cyclooxygenase inhibitor in the apoptosis of prostate carcinoma cells. Methods The human prostate cancer cells PC-3 and LNCaP were routinely cultured. Total RNA was isolated from the cultured cells, normal prostate tissues and benign hyperplasia prostate tissues by using TRIZOL. The RT-PCR reaction was performed to detect the COX-1 mRNA, COX-2 mRNA. COX-2 was analyzed by Western blot in the controlled and treated teamsof the cells. Results Using RT-PCR analysis, we detected the segments of eDNA corresponding to specific COX-1 mRNA and COX-2 mRNA extracted from the specimens from prostate cancer cells, benign hyperplasia tissues, and normal prostate tissues. We detected a slight but clear band of COX-1 mRNA in all samples. However, we only detected a specific band of COX-2 mRNA in the samples from prostate cancer tissues. It is noteworthy that the survival of PC-3 and LNCaP cells was affected by celecoxib in a dose- and time-dependent manner. By Western blot analysis, we found that the level of COX-2 expression in controlled teams of the cells was higher than that of the treated teams of the cells. Conclusions There is a marked expression of cyclooxygenase in prostate carcinoma cell lines prostate cancer cells.
