摘要
目的探讨血必净注射液对百草枯中毒大鼠多脏器损伤的保护作用及其机制。方法Wistar大鼠54只,随机分为对照组(n=6)、染毒组(n=24)、治疗组(n=24)。染毒组和治疗组一次性胃内灌入20%百草枯溶液(50mg/kg),对照组胃内灌入等体积的生理盐水后处死留取标本。染毒后染毒组给予生理盐水4ml/kg腹腔注射,治疗组给予血必净注射液4ml/kg腹腔注射,均每天1次,连续给药7d或至处死。于第1、3、7、14天各取6只大鼠处死,留取血液及肺组织标本。采用硫代巴比妥酸比色法测定大鼠血浆丙二醛(MDA)含量,黄嘌呤氧化酶法测定超氧化物歧化酶(SOD)活力,ELISA法测定血清TNF-α及IL-10含量,肌酸激酶同工酶(CK-MB)、谷丙转氨酶(ALT)、肌酐(Cr)及淀粉酶(AMS)测定由全自动生化分析仪完成。肺组织行HE染色,光镜下观察其病理学变化。结果染毒后染毒组及治疗组MDA均明显升高,SOD明显下降,但治疗组MDA值低于染毒组,SOD值高于染毒组;2组染毒后TNF-α及IL-10均有明显升高,染毒组升高更明显;染毒后染毒组及治疗组CK-MB、ALT、Cr及AMS均有不同程度改变,但治疗组各项指标优于染毒组。光镜下染毒组于染毒后第14天可见肺组织慢性炎症改变,局部区域见纤维组织增生,间质增厚;治疗组炎症改变较轻,未见纤维化征象。结论血必净注射液可通过抗氧化及调控炎症介质作用,减轻多脏器损伤,对百草枯中毒有一定治疗作用。
Objective To explore the protective effect and its potential mechanism of xuebijing injection on multiple organs of rats with paraquat poisoning.Methods Fifty-four male Wistar rats were randomly divided into control group (n=6),exposed group (n=24) and treated group (n=24).Rats in exposed group and treated group were gavaged 20% paraquat (50 mg/kg).Rats in control group were gavaged the same dose saline and taken the specimens after execution.Exposed group were given saline (4 ml/kg) by intraperitoneal injection (IP)after exposed to poison;treated group were given xuebijing injection (4 ml/kg) by IP after exposed to toxicant;IP was administered once a day for 7 days or until to be executed.At day 1,3,7,14 after exposed to toxicant,six rats were executed respectively,blood and lung tissue samples were collected.Malondialdehyde(MDA) content was measured by colorimetry of thiobarbituric acid;superoxide dismutase(SOD) activity was measured by the method of xanthine oxidase;serum tumor necrosis factor(TNF)-α and interleukin(IL)-10 content were determinated by the method of ELISA;creatine kinase isozyme(CK-MB),alanine transarninase(ALT),creatinine (Cr) and amylase (AMS) were determinated by the full-automatic biochemical analyzer.Lung tissue sample was stained by HE,and the pathological changes were observed under microscope.Results Compared with control group,plasma concentration of MDA were significantly increased,and SOD activity were decresed markly after exposed to toxicant in exposed group and treated group.But MDA content was lower,and SOD activity was higher in treated group than those in exposed group.The serum concentration of TNF-α and IL-10 increased significantly in exposed group and treated group,but the increase was more obvious in the exposed group.Levels of CK-MB,ALT,Cr and AMS changed in different degrees after exposed to poison in exposed group,which in the treated group were better.Chronic inflammation,fibrous tissue proliferation in local area and interstitial thickening were found on the 14th day in exposed group,but the treated group only showed mild inflammation and no sign of fibrosis.Conclusions Xuebijing injection may reduce the multiple organ damages possibly by anti-oxidation and regulating the inflammatory mediators,therefore it has an certain therapeutic effect in paraquat poisoning.
