摘要
目的:观察RNA干扰对人食管癌裸鼠移植瘤组织中血管内皮生长因子C(VEGF-C)表达的影响。方法:将转染VEGF-C小分子干扰RNA(siRNA)载体质粒(HX1、HX2和HX3)的EC9706细胞分别注射于裸鼠皮下,以转染无关序列和正常EC9706细胞为对照,SPF环境饲养4周后处死,取移植瘤组织,分别采用RT-PCR和免疫组化SP法检测VEGF-CmRNA和蛋白的表达。结果:5组移植瘤体积、质量及VEGF-C蛋白和mRNA相对表达量差异均有统计学意义(F分别为3.372、3.948、7.621和8.916,P均<0.05)。与正常细胞对照组和无关序列组相比,HX1、HX2及HX3转染组移植瘤组织中VEGF-C蛋白及mRNA表达明显降低(P<0.01)。结论:VEGF-CsiRNA能有效抑制裸鼠食管癌移植瘤组织中VEGF-CmRNA及蛋白的表达。
Aim:To study the effect of RNA interference on expression of vascular endothelial growth factor C(VEGF-C) in human esophageal cancer cell line EC9706 in vivo.Methods:EC9706 cells were allocated into 5 groups and the first 4 groups were transfected with pSINsi-U6-HX1,pSINsi-U6-HX2,pSINsi-U6-HX3,and non-related sequence.Then nude mice received a subcutaneous injection of EC9706 cells from the above groups transfected by siRNA in SPF cube.Mice were killed 4 weeks later.VEGF-C mRNA and protein in the transplanted neoplasm tissue were detected by RT-PCR and immunohistochemistry,respectively.Results:The size and mass of transplanted neoplasm,and the expressions of VEGF-C protein and mRNA among the 5 groups had significant differences(F was 3.372,3.948,7.621 and 8.916,P 0.05).The expressions of VEGF-C protein and mRNA in the groups of pSINsi-U6-HX1,HX2,and HX3 decreased compared with the other 2 groups(P 0.01).Conclusion:The specific siRNA of VEGF-C can inhibit the expression of VEGF-C in EC9706 cells in vivo significantly.The siRNA against VEGF-C might serve as a potential method for cancer treatment in human esophageal cancer.
