1,25-二羟维生素D3对猪成釉细胞釉原蛋白和成釉蛋白分泌的影响

Effect of 1,25-(OH)_2 D_3 on the expression of amelogenin and ameloblastin secreted by porcine ameloblasts

摘要 目的:探讨1,25-二羟维生素D3[1,25-(OH)2D3]对体外培养的猪成釉细胞釉原蛋白(AMEL)及成釉蛋白(AMBN)分泌的影响。方法:原代培养猪成釉细胞,取第2代细胞12瓶,分为4组,每组3瓶。对照组不作加药处理,其余各组分别加入1×10-10、1×10-8及1×10-6mol/L的1,25-(OH)2D3,并分别于加药后1、3及5d取出1瓶,采用Western blot方法检测AMEL及AMBN的表达。结果:猪成釉细胞AMEL和AMBN的表达在不同浓度组间(F=359.688、619.718、117.412、859.364和49.099、705.946、338.824、119.352,P均<0.001)与不同作用时间组间(F=789.133、116.071、196.509和307.327、657.313、762.749,P均<0.001)差异均有统计学意义。其中AMEL的表达随作用时间延长和药物浓度的增加而增强,AMBN的表达随作用时间的延长增强,随药物浓度的增加而减少。结论:1,25-(OH)2D3可能通过调节AMEL及AMBN在釉质中的相对含量影响釉质的矿化过程。 Aim:To investigate the effect of 1,25-dihydroxyvitamin D3[1,25-(OH) 2D3]on amelogenin(AMEL) and ameloblastin(AMBN) secreted by cultured porcine ameloblast.Methods:A totle of 12 bottles of the second generation cultured porcine ameloblast were divided into four groups equally.Each bottle of the group were added 0,1 × 10-10,1 × 10-8,1 × 10-6 mol/L 1,25-(OH) 2D3,respectively.After 1,3 and 5 days treatment,the expression of AMEL and AMBN were detected using Western blot.Results:The expression of AMEL and AMBN in each 1,25-(OH) 2D3 concentration group and each time set were different,and there were obvious significance(F = 359.668,619.718,117.412,859.364,789.133,116.071,196.509,and 49.099,705.946,338.824,119.352,307.327,657.313,762.749,P 0.001).The expression levels of AMEL increased with the time prolonged and the concentration of 1,25-(OH) 2D3 increased.The expression levels of AMBN increased with the time prolonged,while decreased with the concentration of 1,25-(OH) 2D3 increased.Conclusion:1,25-(OH) 2D3 may regulate the relative content of AMEL and AMBN in enamel,thus affect the enamel mineralization.
