摘要
目的:探讨养阴化纤方对博莱霉素致大鼠肺纤维化的作用及机制。方法:选取雄性SD大鼠60只,随机分为正常对照组、模型对照组、阳性对照组、高剂量治疗组和低剂量治疗组5组,每组12只;后4组用盐酸博莱霉素建立大鼠肺纤维化模型,高剂量治疗组和低剂量治疗组分别灌服养阴化纤方药20和10g/kg,阳性对照组灌服等体积的地塞米松,正常对照组灌服等体积的生理盐水;灌药后第7和28天,分别检测大鼠支气管肺泡灌洗液肿瘤坏死因子-α(TNF-α)、内皮素(ET-1)和肺组织羟脯氨酸(HYP)的水平,同时观察肺组织形态学变化。结果:5组间TNF-α、ET-1和HYP水平比较,差异均有统计学意义(第7天:F值分别为36.823、12.064和51.979,P均<0.001;第28天:F值分别为23.484、19.600和33.631,P均<0.001)。且模型对照组的TNF-α、ET-1和HYP水平均高于正常对照组(P均<0.05),阳性对照组、高剂量治疗组和低剂量治疗组的TNF-α、ET-1和HYP水平均低于模型对照组(P均<0.05),肺纤维化程度亦低于模型对照组。结论:养阴化纤方可能是通过降低肺组织TNF-α、ET-1和HYP的水平而达到防治肺纤维化的效应。
Aim:To study the mechanism of Yangyin huaxian prescription on the rats models with pulmonary fibrosis.Methods:Sixty SD male rats were randomly divided into five groups:normal group,model group,high-dose treatment group,low-dose treatment group,and prednisone group(n = 12).The experimental pulmonary fibrosis models were produced by injecting BLMA5.The drugs were given daily starting from the second day and the rats were killed on the 7th and 28th day.The shape of rat lung tissues was observed and the levels of TNF-α,ET-1 in BALF and hydroxyproline(HYP) in lung tissues were measured.Results:There were significant differences in levels of TNF-α,ET-1 and HYP among five groups(7th day:F was 36.823,12.064,and 51.979,P〈0.001;28th day:F was 23.484,19.600,and 33.631,P〈0.001).The levels of TNF-α,ET-1 and HYP in model group were significantly higher compared with those in normal group(P〈0.05).The levels of TNF-α,ET-1 and HYP in high-dose treatment group,low-dose treatment group and prednisone group were significantly higher compared with those in model group(P〈0.05).The grade of collagen deposition of high-dose treatment group,low-dose treatment group and prednisone group was lower than that of model group obviously.Conclusion:Yangyin huaxian prescription may inhibit the pulmonary fibrosis by decreasing the levels of TNF-α,ET-1 and HYP.
