摘要
目的研究C5-siRNA对大鼠心肌缺血损伤的血流动力学的影响。方法 30只Sprague-Dawley(SD)大鼠随机分为假手术组,对照组和C5-siRNA干预组。结扎对照组和干预组大鼠的冠状动脉(以下简称冠脉)前降支近端制备急性大鼠心肌缺血模型,在冠脉前降支支配的中心区分别注射0.9%氯化钠液和C5-siRNA,1h后松扎,测定血流动力学指标及血清乳酸脱氢酶(LDH)和肌酸激酶(CK-MB)变化。结果与模型组比较,C5-siRNA对心肌缺血大鼠血流动力学指标均有一定的改善趋势,尤其对左心室收缩压(LVSP)、±dp/dtmax等指标在某些时间点影响显著(P<0.05),C5-siRNA对心率(HR)影响则不明显。C5-siRNA对严重的心肌缺血引起的血清LDH和CK-MB同工酶升高有明显降低作用,尤其是与模型组CK-MB相比(P<0.05)。结论 C5-siRNA对大鼠急性心肌缺血具有保护作用,对心肌缺血大鼠血流动力学指标有一定的改善趋势。
Objective To study the impact of C5-siRNA on the hemodynamics status of acute myocardial ischemia rats.Methods Thirty healthy Sprague-Dawley rats were randomly divided into sham,model and C5-siRNA groups.The rat of model and C5-siRNA groups were established by ligating the left anterior descending coronary artery.The rats were infused with 100 μl/kg of C5-siRNA into vein in C5-siRNA group and equal quantity of normal saline in ischemia-reperfusion (IR) group after ligating the LAD 20 min for 30 min and then performing reperfusion for 120 min.The parameter of dynamics,LDH and CK-MB in serum were measured.Results C5-siRNA improved the parameter of dynamics,such as the left ventricular systolic pressure (LVSP),the left ventricular maximum pressure rate of change (± dp/dtmax),and the left ventricular end-diastolic pressure (LVEDP),and there was significant difference at some time(compare with model group,P0.05),but no impact on heart rate.Compare with model group,serum LDH and CK-MB in C5-siRNA group were diminished significantly(P0.05).Conclusions C5-siRNA may improve the parameter of dynamics and protect the rats suffering from acute myocardial ischemia.
