丹酚酸B作用不同时间对高糖高脂内皮细胞凋亡的抑制作用

摘要 目的观察丹酚酸B抑制高糖高脂诱导的内皮细胞凋亡的作用。方法以糖基化终末产物(AGEs)和氧化低密度脂蛋白(ox-LDL)联合作用诱导人脐静脉内皮细胞凋亡,通过Hochest33258染色,观察并比较丹酚酸B及洛伐他汀作用不同时间(12、24、36、48、72h),抑制高糖高脂诱导的内皮细胞凋亡的效果。结果研究表明,洛伐他汀作用12、24h时,抑制内皮细胞凋亡效果明显,而丹酚酸B作用36、48h及72h抑制内皮细胞凋亡效果显著。结论与洛伐他汀相比,丹酚酸B抑制高糖高脂诱导的内皮细胞凋亡的作用发挥得较迟缓,但效果持久。 Objective To compare the effects of salvianolic acid B and lovastatin inhibiting endothelial cells apoptosis after incubation for different time.Methods Human umbilical vein endothelial cells (HUVECs) apoptosis were induced by AGEs and ox-LDL.After HUVECs were incubated salvianolic acid B and lovastatin for 12,24,36,48,72 h,percentage of apoptosis were observed under fluorescent microscope by Hochest 33258 staining.Results Although treatment with lovastatin for 12,24 h,apoptosis decreased significantly (P0.05),treatment with salvianolic acid B for 36,48 and 72 h,apoptosis decreased significantly (P0.05).Conclusions Compared with lovastatin,salvianolic acid B might exert a later but lasting inhibition effect on endothelial cell apoptosis induced by high-sugar and high-fat.
