摘要
目的:了解不同浓度胰岛素对毛乳头细胞的生物学作用。方法:人头皮组织分离、培养、获得毛乳头细胞,根据胰岛素浓度分为1×10-8、1×10-7、1×10-6mol/L3个组,设无胰岛素培养液为对照组。应用不同浓度胰岛素的2%新生牛血清培养液培养毛乳头细胞3天,分别用MTT法测定毛乳头细胞的增殖;AnexinV/PI双染色法测定毛乳头细胞的凋亡情况;体外细胞划痕法测定其迁移能力。结果:与对照组比较,1×10-8、1×10-7mol/L胰岛素组毛乳头细胞增殖、抗凋亡及迁移能力显著增加(P<0.05或P<0.01),且1×10-7mol/L组作用最强(P<0.05或P<0.01)。结论:1×10-8~1×10-7mol/L胰岛素可以增强毛乳头细胞增殖、迁移和抑制凋亡的作用。
Objective To study the effects of different concentration insulin on a papillose cell.Methods Papillose cell was isolated from human scalp dermal tissue and cultured.Then to use 2% new-born calf serum of different concentration insulin(1×10^-8,1×10^-7,1×10^-6 mol/L) culturing papillose cell for 3 days.And blank control group was set up.Cells counting by the MTT method to determined the proliferation;Annexin Ⅴ/PI was used to investigate the apoptosis rate of papillose cel,the cell migration was measured by in intro wound-healing assay.Results Compared with those in control group,the 10^-8、10^-7mol/L insulin groups significantly promote the proliferation,migration while suppress apoptosis of papillose cell than that in control group and the 10^-6 mol/L insulin group(P〈0.05 or P〈0.01),and the 1×10^-7 mol/L group was the best one(P〈0.05 or P〈0.01).Conclusion The concentration of 10^-8~10^-7 mol/L insulin could significantly promote the proliferation,migration while suppress apoptosis of papillose cell.
