摘要
目的:通过对葡萄干中黄曲霉毒素(G2、G1、B2、B1)的高效液相色谱柱后衍生和荧光计两种不同方法测定比较,从而获得准确、可靠的检测结果。方法:样品经甲醇水溶液提取,经免疫亲和柱层析净化,甲醇洗脱,分别按操作要求进行高效液相色谱柱后衍生和荧光计两种不同方法测定。结果:采用荧光计法对葡萄干样品进行检测,结果基本均呈显阳性(样品本身存在有较强的荧光干扰),应采用高效液相色谱柱后衍生进一步确认,方可获得准确、可靠的结果。结论:荧光计法检测出呈阳性结果的样品常伴有假阳性,因此对存在干扰的样品,可直接用高效液相色谱柱后衍生进行检测,既省时又提高效率,而对筛查大量样品结果,荧光计法则起了一定的快速检测作用,若是阴性结果,则不必高效液相色谱柱后衍生进一步确认。
Objective:Through after raisin in Aflatoxin(G2,G1,B2,B1) highly effective liquid phase chromatographic column derivation and fluorimeter two different methods determination comparison,thus obtains accurately,the reliable examination result.Methods: The sample after the methyl alcohol water-soluble fluid extraction,kisses after the immunity with the column chromatographic analysis purifies,the methyl alcohol elution,after presses the operation request to carry on the highly effective liquid phase chromatographic column separately the derivation and the fluorimeter two different method determinations.Results: Uses the fluorimeter law to carry on the examination basically to the raisin sample,the result assumes reveals the masculine gender(sample itself to exist has strong fluorescence disturbance),after should use the highly effective liquid phase chromatographic column to grow further confirmed,only then obtains accurately,the reliable result.Conclusion: The fluorimeter law examines assumes the masculine result the sample often to be accompanied by the false positive,therefore to has the disturbance the sample,may grow directly after the highly effective liquid phase chromatographic column carries on the examination,both time-saving and enhances the efficiency,but to sieved looks up the massive sample result,the fluorimeter principle played certain fast examination role,if negative result,after then did not need the highly effective liquid phase chromatographic column to grow further confirmed.
