尼古丁诱导Ref-1磷酸化抑制DNA损伤的修复

摘要 目的研究尼古丁抑制DNA损伤后修复的分子机制。方法 Western blot检测蛋白表达水平,同位素方法检测Ref-1磷酸化和其核酸内切酶活性,试剂盒定量检测AP位点。结果尼古丁可以诱导Ref-1磷酸化,下调其核酸内切酶活性,抑制AP位点的修复,星状孢子素(Staurospo rine)和2-(2-氨基-3-甲氧基苯基)-4H-1-苯并吡喃-4-酮(PD98059)可以抑制尼古丁诱导的Ref-1磷酸化。结论尼古丁通过诱导Ref-1磷酸化,下调其核酸内切酶活性,抑制DNA损伤的修复。 【Objective】 To study the molecular mechanism of Nicotine inhibiting DNA damage repair.【Methods】Detecting the level of protein expression by Western blotting.Detecting the Ref-1 phosphorylation and activity of its endonuclease by isotope method.Detecting AP sites by DNA damage kit.【Results】Nicotine induced Ref-1 phosphorylation,down-regulated its endonuclease activity and inhibited AP sites repair.Staurosporine and PD98059 inhibited Ref-1 phosphorylation induced by Nicotine.【Conclusion】Nicotine can down-regulate endonuclease activity of Ref-1 and inhibit DNA damage repair by inducing Ref-1 phosphorylation.
