全反式维甲酸抑制NDRG1基因在宫颈癌HeLa细胞中的表达

摘要 目的观察全反式维甲酸对人宫颈癌HeLa细胞体外生长的影响;探讨全反式维甲酸对人宫颈癌HeLa细胞影响的机制及其与NDRG1基因的关系;为全反式维甲酸应用于宫颈癌的治疗提供实验依据。方法用不同浓度全反式维甲酸作用于人宫颈癌HeLa细胞,采用MTT比色法检测细胞生长情况;RT—PCIR和Westernblot检测全反式维甲酸对宫颈癌HeLa细胞中NDRG1基因和蛋白表达的影响。结果全反式维甲酸能够抑制宫颈癌HeLa细胞生长,并且这种抑制作用具有浓度和时间的依赖性(P〈0.05);全反式维甲酸能够上调宫颈癌HeLa细胞中NDRG1基因和蛋白的表达。结论全反式维甲酸能够抑制宫颈癌HeLa细胞生长,这种抑制作用与NDRG1的表达上调相关。
