布托啡诺对大鼠κ-阿片受体mRNA表达的影响

Effects of butorphanol on mRNA expression of κ opiate receptor in rats

目的观察鞘内注射布托啡诺对大鼠中枢神经系统κ-阿片受体mRNA(κ-mRNA)表达的影响。方法 96只成年雄性SD大鼠随机分为布托啡诺10、20、30μg组(L、M、H组)和生理盐水组(N组),每组24只。观察鞘内注射不同剂量布托啡诺或生理盐水后甩尾潜伏期(TFL)的变化,检测脊髓、脑干及脑皮质κ-mRNA的表达。结果鞘内注射布托啡诺具有镇痛作用,并呈时间和剂量依赖性地增加脊髓、脑干、脑皮质中的κ-mRNA的表达。结论鞘内注射布托啡诺可促进大鼠脊髓、脑皮质和脑干κ-mRNA的表达。

Objective To investigate the effect of butorphanol on expression of κ opiate receptor in central venous system in rats.Methods Ninety-six SD rats were randomly divided into 4 groups with 24 rats each.The rats in groups of L,M,H were administered intrathecally butorphanol 10 μg,20 μg and 30 μg,respectively.Normal saline was given in group NS as the control.The time of rat tail-flick latency（TFL） was recorded.The κ-mRNA expressions of the spinal cord,brainstem and cerebral cortex were determined at 1,2 and 4 hours later.Results Butorphanol injected intrathecally had analgesic effect.The κmRNA expressions in spinal cord,brainstem and cerebral cortex were significantly increased in a time-and dose-dependent manner in groups of L,M and H,which were more than those in group NS（P〈0.05） Conclusion Butorphanol can increase the mRNA expression of κ opiate receptor in spinal cord,brainstem and cerebral cortex in rats.
