血府逐瘀汤对内皮祖细胞功能的影响 被引量:15

摘要 目的:观察血府逐瘀汤对骨髓内皮祖细胞功能的影响。方法:用不同浓度的血府逐瘀汤含药血清和空白对照血清诱导处理内皮祖细胞后,采用MTT、Boyden小室、黏附和吞噬实验观察药物对EPCs增殖、迁移、黏附和吞噬能力的影响。结果:与空白对照组比较,24h15%浓度,48h10%浓度的含药血清对EPCs增殖有明显促进作用;10%和15%含药血清作用3个时间段对EPCs的迁移和黏附功能均有显著的促进作用,但5%含药血清影响黏附功能的作用从抑制到促进;10%和15%药物作用48h均可影响EPCs的吞噬功能,诱导时间延长至72h,只有10%浓度有显著性差异。结论:血府逐瘀汤能显著影响内皮祖细胞增殖、迁移、黏附和吞噬ac-LDL的能力,提示其具有诱导内皮祖细胞转化成内皮细胞参与血管新生的作用。 Objective :To observe the effect on EPCs function by Xuefu Zhuyu Decoction. Methods :After induced by serial concentrations of Xuefu Zhuyu Decoction-contained serum(XZDCS)and blank serum,EPCs proliferation,migration,adhension and uptake function were detected by MTT,Boyden chamber,adhension and uptake of ac-LDL respectively. Results :Compared with the control group,15% and 10% XZDCS could elevate the cell regeneration for 24 h and 48 h respectively. Both concentrations could improve the EPCs migration and adhension for all 24,48,72 h and uptake of ac-LDL only 48 h. But 10% XZDCS could last effect on uptake of ac-LDL to 72 h and 5% XZDCS converted the inhibition of cell adhension in the first 24 h to promotion in the next 48-72 h. Conclusion :Xuefu Zhuyu Decoction could induce EPCs differentiation into EC to angiogenesis by promoting its proliferation,migration,adhension and uptake function.
