环孢霉素A治疗哮喘的实验研究

EXPERIMENTAL STUDY OF THE THERAPEUTIC EFFECT OF CYCLOSPORIN A ON ASTHMA

目的：探讨环孢霉素Ａ对哮喘的治疗作用及机制。方法：２１只豚鼠随机分为正常组、模型组及环孢霉素Ａ组。用卵白蛋白制作豚鼠哮喘模型。用环孢霉素Ａ２５ｍｇ／（ｋｇ·ｄ）连续腹腔注射３ｄ，检测外周血Ｔ淋巴细胞增殖反应，右侧肺脏做支气管肺泡灌洗液中细胞成分分析，左侧肺脏做组织学检查。结果：环孢霉素Ａ组Ｔ细胞增殖刺激指数显著低于模型组（Ｐ＜０．０１）；支气管肺泡灌洗液中细胞总数、嗜酸性粒细胞计数也显著低于模型组（Ｐ＜０．０１）；气道壁炎症反应减轻；且刺激指数与支气管肺泡灌洗液中嗜酸性粒细胞计数之间呈显著正相关。结论：环孢霉素Ａ通过抑制Ｔ细胞活化增殖抑制气道炎症。

OBJECTIVE:To study the therapeutic effect of cyclosporin A on asthma in guinea pigs.METHODS:The model of asthma was established.The T lymphocyte proliferative response was measured,inflammatory response in the airway was assessed by both examination of the cells of bronchoalveolar lavage fluid and the lung histology.RESULTS:The total cell number,the eosinophil number in bronchoalveolar lavage fluid of cyclosporin A treated group were significantly lower than those of the model group.The stimulation index of T lymphocyte proliferative respose of cyclosporin A treated group was also significantly lower than that of the model group.The eosinophil infiltration within airway walls was markedly inhibited compared with the model group.The stimulation index was positively correlated with the number of eosinophil in bronchoalveolar lavage fluid.CONCLUSION:Cyclosporin A can inhibit the eosinophil infiltration within airway walls by suppressing T cell proliferation and activation;cyclosporin A have potential therapeutic effect for asthma.
