摘要
衰变加速因子(DAF)是补体激活途径中的一个重要调节蛋白,转人DAF(hDAF)基因猪可以作为异种器官移植的供体.采用直接免疫荧光法对转hDAF基因猪耳组织冰冻切片进行了观察,并应多媒体图文分析系统测定荧光照片的平均灰度.结果表明,14头转基因猪中,有7头动脉肌肉展表达了外源hDAF基因,它可以作为外源基因在供体中表达的主要指标,用于转基因猪的检测.
Abstract: Decay accelerating factor(DAF) is one of the important membrane regulating protein in the complemcnt activating ways. The transgenic pigs for human DAF gene can used for xenotransplatationdonors. Direct immunofluorescent was used to study ear biopsy specimens of transgenic pigs and mutimediacomputer picture assay system was used to measure average fluorescent photos.Of 14 transgenic pigs, wefound 7 transgenic pigs of which vascular smooth muscle expressed exogene of hDAF and it could be con-sidered an excellent marker.
