氯喹对烟雾吸入伤大鼠肺细胞膜ATP酶活性和丙二醛含量的影响 被引量:1

摘要 目的 :探讨氯喹对烟雾吸入伤大鼠肺细胞膜ATP酶活性及丙二醛含量的影响。方法 :80只Wistar大鼠随机分成正常对照组 ,烟雾吸入伤 1 ,3 ,6,1 2和 2 4h组以及氯喹治疗 6h和 1 2h组 ;分别于各时相点活杀动物 ,取肺制备膜制剂 ,用生化比色法测定膜上Na+ ,K+ -ATP酶、Mg2 + -ATP酶和Ca2 + -ATP酶活性 ,用比色法测定膜上丙二醛含量 ,并用定磷法测定膜总磷脂含量。结果 :烟雾吸入伤后 ,肺细胞膜 3种ATP酶活性从伤后 1h至伤后 2 4h均明显降低 (P <0 .0 1 ) ,同时伴有膜丙二醛含量增加和膜总磷脂含量降低 ;氯喹治疗 6h组Na+ ,K+ -ATP酶、Ca2 + -ATP酶活性和总磷脂含量明显高于吸入伤 6h组 ,而丙二醛却明显低于吸入伤 6h组 (P <0 .0 5与P <0 .0 1 )。结论 :肺细胞膜磷脂分解、丙二醛含量增加和ATP酶活性降低可能是烟雾吸入伤发生发展的重要原因 ;氯喹部分改善ATP酶活性、降低丙二醛含量。 OBJECTIVE:To explore the influences of chloroquine on the activities of adenosinetriphosphatases(ATPases) and the content of malondialdehyde(MDA) on rats model of smoke inhalation injury.METHODS:Eighty wistar rats were randomly divided into normal control group,injurious groups at 1,3,6,12 and 24h following smoke inhalation,and chloroquine treatment groups(chloroquine was purchased from Sigma,10mg/kg,immediatly injected intraperitoneally after smoke inhalation) at 6h and 12h following smoke inhalation.The rats were killed at each time site and the lungs were incised for preparing pulmonary cellular membrane,the activities of Na +,K +-ATPase,Mg 2+ -ATPase and Ca 2+ -ATPase were detected by biochemistry-chromatometric method,the content of MDA was assayed by chromatometric method and the content of total phosphatide of pulmonary cellular membrane was measured by quantified phosphorus.RESULTS:The activities of three kinds of ATPases were declined significantly from 1 hour to 24 hours after smoke inhalation injury(P<0.01),with increasing in the content of MDA and decreasing in the content of total phosphatide of pulmonary cellular membrane,but the activities of Na +,K +-ATPase and Ca 2+ -ATPase and the content of total phosphatide were increased and the content of MDA was decreased in 6h group of chloroquine treatment while comparing with 6h group of smoke inhalation injury(P<0.05).CONCLUSION:The decomposition of phosphatide,the increase in the content of MDA and the decreases in the activities of ATPases in pulmonary cellular membrane might be important factors of the occurrence and the development of smoke inhalation injury.Chloroquine might prevent and treat smoke inhalation injury for partially improving the activities of ATPases,decreasing the content of MDA of pulmonary cellular membrane and stabilizing pulmonary cellular membrane.
