增龄对大鼠肾缺血再灌注肾小管上皮细胞凋亡的影响

摘要 采用原位末端转移酶标记技术(TUNEL)标记法,结合肾组织切片HE染色,观察并比较3个月与24个月大鼠肾缺血再灌注不同时相损伤所激发的细胞凋亡现象。结果发现肾缺血再灌注12、48h后,各年龄组大鼠肾小管上皮细胞凋亡水平均明显高于正常对照大鼠(P<0.001);且于正常和缺血再灌注各时相组,24月龄组的肾小管上皮细胞凋亡水平均明显高于3月龄大鼠(P<0.01)。提示肾小管上皮细胞凋亡是缺血再灌注后肾小管细胞死亡的一种主要方式,随年龄增长机体可通过细胞凋亡的调节机制增加细胞凋亡水平,达到清除损伤。 The effect of aging on apoptotic level of renal tubular cells during reperfusion after acute renal ischemia in SD rats was studied. A model of bilateral post ischemic renal injury by clamping renal pedicles for 45 minutes was setup. The levels of apoptosis in rats (3, 24 months old, respectively) were observed and compared. In this process of analysis, TdT mediated dUTP nick and labeling (TUNEL) technique and the renal tissular HE staining were used. The result showed that the apoptotic levels in 3, 24 months old rats after 12, 48 hours of post ischemic reperfusion were significantly higher than those in normal control rats ( P <0.001). The apoptotic level in 24 months old rats, however, was remarkably higher than that in 3 months old rats in both the normal control group and the groups of post ischemic reperfusion ( P<0.001 ). It was concluded that renal tubular apoptosis was the main way of death after renal post ischemic reperfusion. The level of apoptosis could be improved so as to clear the injuried and deteriorated cells by the mechanisms of regulating apoptosis with aging.
