阿糖胞苷、干扰素对慢性粒细胞白血病细胞凋亡影响的研究 被引量:2

摘要 将31例慢性粒细胞白血病(CML)患者的骨髓单个核细胞(MNC)在体外与干扰素、阿糖胞苷作用一定时间后,以AO/EB法和FCM法检测细胞凋亡率。结果:①干扰素、阿糖胞苷均可诱导CML细胞凋亡,前者诱导凋亡的强度强于后者(P<0.01)。②干扰素诱导凋亡,在CML不同病期间差异显著(P<0.01)。③阿糖胞苷与干扰素有协同诱导凋亡作用。④干扰素体外诱导凋亡率的高低与临床治疗效果密切相关。提示:诱导凋亡是干扰素、阿糖胞苷治疗CML的一个机制。 Different concentrations of IFN α and Ara c were added to MNC from bone marrow of 31 CML patients. Two methods, AO/EB and FCM, were used in quantitative detection of cell apoptosis. The results demonstrated: ①Both IFN α and Ara c can induce apoptosis of CML cells, and the effect of the former was stronger than that of the latter ( P <0.01). ②When IFN α was used to induce CML cell apoptosis, significant difference among the different phase groups was found ( P <0.01). ③IFN α and Ara c acted synergistically in inducing cell apoptosis. ④Positive correlation between the in vitro and in vivo studies was found. It was suggested that IFN α and Ara c may affect CML cells by inducing cell apoptosis, and to induce cell apoptosis in vitro can be used as an effective method in selecting suitable CML patients treated with IFN α.
