摘要
目的:探讨丙丁酚(普罗布考)对氧化低密度脂蛋白(oxLDL)诱导人脐静脉血管内皮细胞(HUVEC)损伤的保护作用,以及对NF-κB和IκB-α表达的调节作用。方法:MTT法分别检测oxLDL不同浓度(1g/ml、10g/ml、100g/ml)处理HUVEC不同时间(12h、24h、48h)细胞增殖情况;5g/mL丙丁酚预处理HUVEC24小时,MTT检测细胞增殖情况;Western blot检测oxLDL处理细胞及丙丁酚预处理后NF-κB和IκB-α蛋白表达水平。结果:oxLDL处理HUVEC细胞24小时后,对细胞增殖具有抑制作用,Western blot结果显示NF-κB表达显著上调,IκB-α蛋白表达下调。而用5ug/mL丙丁酚预处理可以明显缓解细胞增殖抑制,NF-κB和IκB-α蛋白的表达也恢复至正常水平。结论:丙丁酚可显著缓解oxLDL诱导的HUVEC细胞增殖抑制,并且NF-κB和IκB-α表达的改变可能参与此过程。
Objective:To investigate the protect of oxLDL on oxLDL-induced inhibition of HUVEC cell proliferation, and explore the regulation of NF-κB and IκB-α.Methods:Pretreated the HUVEC with probucol in different dose (1 g/mL, 10 g/mL, 100 g/mL) and time(12h, 24h, 48h), and proliferation of cells were examined by MTT;then 5 g/mL probucol pretreated the HUVEC for24h, MTT method measured the proliferation of cells.NF-κB and IκB-α protein were measured by Western blotting after treated with probucol.Results:The cell proliferation was significantly inhibited when treated with probucol.The result of Western blotting showed that NF-κB was down-regulated, IκB-α was up-regulated.While 5ug/mL probucol pretreated the HUVEC, the inhibiation of proliferation relieved, and the expression of NF-κB and IκB-α returned to normal levels.Conclusions:Probucol significantly relieve the oxLDL-induced inhibition of HUVEC cell proliferation, and the alterant expression of NF-κB and IκB-α may be involved in this process.
