哮喘大鼠肾上腺皮质功能变化与白细胞介素10水平的关系

Relationship between the alterations of adrenal cortical function and IL-10 level in asthmatic rats

摘要 目的探讨哮喘大鼠肾上腺皮质功能变化与白细胞介素10(IL-10)水平的关系。方法卵蛋白致敏方法制备大鼠哮喘模型,随机分为哮喘急性发作期组(A组)、哮喘缓解期组(B组)、正常对照组(C组)、模型对照组(D组),每组6只。放射免疫分析法测定血浆皮质酮(CORT)、促肾上腺皮质激素(ACTH)以及下丘脑促肾上腺皮质激素释放激素(CRH)的含量;ELASA法检测下丘脑组织和血浆IL-10的水平。结果与C组比较,A组大鼠脑组织和血浆IL-10水平均下降[(15.45±4.06)pg.mg-1.prot-1vs.(28.74±8.91)pg.mg-1.prot-1,和(26.62±6.78)pg/mlvs.(54.86±14.32)pg/ml](P<0.01);CORT、ACTH以及CRH的含量降低[(1.47±0.11)ng.mg-1.prot-1vs.(1.84±0.12)pg.mg-1.prot-1,(75.55±8.17)pg/mlvs.(108.34±16.57)pg/ml,和(121.69±17.65)ng/mlvs.(162.01±21.21)ng/ml](P<0.01)。哮喘缓解期大鼠脑组织IL-10水平上升,CRH的含量增高。结论哮喘大鼠存在肾上腺皮质功能减退可能与下丘脑和血中IL-10水平变化有关。 Objective To study the relationship between the alterations of adrenal cortical function and the interleukin-10(IL-10) level in asthmatic rats.Methods Twenty-four rats were randomly divided into one normal control group(C) and three asthma model groups of A,B and D with 6 rats each.The asthma models of rats were established by intraperitoneal injection of ovalbumin.The rats of group A were with acute attack of asthma,those of group B with remission of asthma.Group D was taken as the model control.Plasma corticosterone(CORT) and adrenoeorticotropic hormone (ACTH) and corticotrophin-releasing hormone(CRH) in the hypothalamus tissue were detected by radioimmunoassay.The levels of IL-10 in hypothalamus tissue and plasma were quantified by a double-ligand-linked immunosorbent assay.Results The levels of IL-10 in hypothalamus tissue and plasma were lower in group A than those in group C [(15.45±4.06) pg·mg^-1 ·prot^-1 vs. (28.74±8.91) pg·mg^-1 ·prot^-1 ,and (26.62±6.78) pg/ml vs.(54.86±14.32) pg/ml] (P〈0.01).So did the levels of CORT,ACTH and CRH [(1.47±0.11) ng·mg^-1 ·prot^-1 vs.(1.84±0.12) pg·mg^-1 ·prot^-1 ,(75.55±8.17) pg/ml vs. (108.34±16.57) pg/ml,and (121.69±17.65) ng/ml vs.(162.01±21.21) ng/ml](P〈0.01).The levels of IL-10 and CRH in the hypothalamus were increased significantly in group B.Conclusion The decreases of adrenal cortical function in asthmatic rats may be associated with the down-regulation of IL-10 levels in the hypothalamus and blood.
