摘要 目的:研究二氯化钴(CoCl2)诱导化学缺氧对人牙髓细胞(dental pulp cells,DPCs)增殖和迁移的影响。方法:改良组织块法体外培养获得的人DPCs,通过CoCl2化学模拟缺氧作用后,采用MTT法检测化学缺氧对人DPCs增殖的影响,Transwell法观察化学缺氧对人DPCs迁移能力的影响。结果:CoCl2诱导的化学缺氧可抑制人DPCs的增殖,并呈浓度依赖性;同时对DPCs的迁移有抑制作用。结论:CoCl2诱导化学缺氧能抑制DPCs的增殖和迁移。 AIM:To investigate the influence of cobalt chloride-induced hypoxia on the proliferation and transmigration of human dental pulp cells(DPCs).METHODS:Human DPCs were obtained using modified tissue explant technique in vitro,and then cultured in the environment of mimic hypoxia induced by cobalt chloride.DPC proliferation was measured by methylthiazol tetrazolium(MTT) assay.Transwell method was applied to evaluate the effect of mimic hypoxia on the transmigration of DPCs.The data was statistically analyzed with SPSS11.5 software package.RESULTS:The proliferation ability of DPCs decreased when exposed to cobalt chloride-induced hypoxia in a dose dependent manner.And the transmigration of DPCs was inhibited in hypoxia.CONCLUSION:Cobalt chloride-induced hypoxia inhibits the proliferation and transmigration of DPCs.
