体内外高糖对血管生成素样蛋白3表达的影响

摘要 目的:观察人体血液中葡萄糖浓度对血管生成素样蛋白3(ANGPTL3)基因及蛋白质表达的影响,探讨ANGPTL3与糖尿病发生的关系。方法:以人血清和L02细胞作为研究对象,从临床采集健康人(健康对照组)、高血糖组空腹血清各10例,各组男女比例均为5∶5。用ELISA法测定血清ANGPTL3蛋白质浓度、全自动生化仪测定血糖浓度。将L02细胞置于含5.6、7.1、11.2及20.0mmol/L葡萄糖培养基中培养,以葡萄糖浓度5.6mmol/L为对照组,用RT-PCR方法检测L02细胞ANGPTL3的mRNA表达量,并用Western Blot方法检测其蛋白质表达量。结果:高血糖组血清ANGPTL3蛋白质含量较健康组明显升高,差异有统计学意义(P<0.05)。体外培养基中高浓度葡萄糖可以促进L02细胞ANGPTL3基因和蛋白质表达,与对照组比较差异有统计学意义(P<0.05)。结论:体内外高糖环境均可以上调ANGPTL3的表达。ANGPTL3高表达可能与糖尿病发生存在一定关联。 Objective:To observe the effects of high glucose on angiopioetin-like protein 3 (ANGPTL3) expression in vivo and in vitro,and the relationship between ANGPTL3 and the diabetes mellitus (DM) thereof. Methods:The human serum and cell line L02 were taken as research subjects, and 10 cases of healthy human serum were taken as control group(male: female= 5∶5)and another 10 cases of hyperglycemia serum as experimental group (male∶female=5∶5). L02 cells were cultured in mediums containing 5.6,7.1,11.2 and 20 mmol/L glucose separately,among which 5.6 mmol / L glucose group was taken as control. The relative level of ANGPTL3 protein in serum was measured by ELISA. The relative level of ANGPTL3 mRNA expression was detected by reverse transcription-polymerase chain reaction (RT-PCR). The relative level of ANGPTL3 protein in L02 was measured by Western Blot. Results: The concentration of ANGPTL3 protein was obviously higher in hyperglycemia group than that of control group (P 0.05). The higher concentration of glucose in medium could significantly stimulate the expression of ANGPTL3 mRNA and protein in cultured L02 cells (P 0.05). Conclusion: The higher glucose can upregulate ANGPTL3 expression both in vitro and in vivo. The expression of ANGPTL3 might be related to DM.
