泛素连接酶PIRH2和p53蛋白在胃癌中的表达关系及意义

The significance of expression of ubiquitin ligase PIRH2 and p53 protein in gastric carcinoma

摘要 目的研究PIRH2和p53蛋白在胃癌组织中的表达水平及其与胃癌生物学行为和预后的关系。方法应用免疫组化方法检测84例胃癌组织中PIRH2和p53蛋白的表达水平。结果 PIRH2和p53蛋白表达之间呈显著负相关(P<0.05)。PIRH2和p53蛋白表达水平与胃癌淋巴结转移、远处转移和pTNM分期均密切相关(P<0.05),而与性别、年龄、肿瘤大小、肿瘤部位、组织类型、浸润深度无显著相关(P>0.05)。PIRH2+/p53-组的胃癌患者3年生存率显著低于其他组(PIRH2-/p53-,PIRH2+/p53+,PIRH2-/p53+)(P=0.002,0.013,0.005)。且多因素生存分析显示,PIRH2和p53蛋白水平是独立的预后因素(P=0.003)。结论 PIRH2和p53蛋白表达与胃癌淋巴结转移及预后有关;PIRH2蛋白表达阳性和p53蛋白表达阴性提示胃癌不良预后。 Objective To study the expression level of PIRH2 and p53 protein in gastric cancer tissues and its relationship with biological behavior of gastric cancer and prognosis.Methods The immunohistochemical method was used to detect PIRH2 and p53 protein expression in gastric carcinoma of 84 patients.Results PIRH2 expression was significantly negatively correlated to p53 protein expression(P0.05).The expression levels of PIRH2 and p53 protein were closely related to lymph node metastasis,distant metastasis and pTNM stage(P0.05),but had nothing to do with sex,age,tumor size,tumor location,histological type and depth of invasion(P0.05).The 3-year survival rate of PIRH2+/p53-gastric cancer group was significantly lower than other groups(PIRH2-/p53-,PIRH2+/p53+,PIRH2-/p53+)(P=0.002,0.013,0.005).Multivariate survival analysis showed that PIRH2 and p53 protein levels were independent prognostic factors(P=0.003).Conclusion The expression of PIRH2 and P53 protein is related to lymph node metastasis and prognosis.The positive expression of PIRH2 and the negative expression of p53 predict poor prognosis of gastric cancer.
