天然产物Genistein抑制人胃癌细胞的磷酸化蛋白组学研究

摘要 研究天然产物genistein调控的磷酸化蛋白质分子,探讨其抑癌的作用机理。文章用40μmol/Lgenistein作用于人胃癌细胞SGC-7901后,采用二氧化钛富集磷酸化肽段,利用LTQ-orbitrap质谱进行磷酸化肽段和磷酸化位点鉴定,结果显示在人胃癌细胞SGC-7901和用genistein处理的胃癌细胞SGC-7901共出现45个差异磷酸化蛋白,通过对这些差异磷酸化蛋白质的比较,从蛋白组学研究方面揭示天然产物genistein作用机理。 The research of phosphorylation protein molecules regulated by natural products genistein was to explore its role in the tumor suppressor mechanism. In this paper, 40umol/L genistein was applied on human gastric cancer cell SGC- 7901 following the introduction of phospho-peptide enrichment kit to enrichment of phosphorylated peptides. Phosphorylated peptides and phosphorylation sites were identified using LTQ- orbitrap mass spectrometry. Results showed that there were 45 different phosphorylated proteins between normal human gastric cancer cell SGC- 7901 and the genisteintreatedgastric cancer cell SGC- 7901. Compared with the different phosphorylated proteins, it revealed the mechanism of natural products genistein in the proteomics research.
