牛膝多糖对2型糖尿病大鼠脂肪分化相关蛋白表达的影响 被引量:4

Effect of Achyranthes Bidentata Polysaccharides on Expression of Adipose Differentiation-Related Protein Gene in Type 2 Diabetes Mellitus Rats

摘要 探讨牛膝多糖(Achyranthes bidentata polysaccharides,ABPS)对2型糖尿病(2-DM)大鼠胰岛素抵抗和脂肪分化相关蛋白(Adpose differentiation-related protein,ADRP)基因表达的影响。将Wistar大鼠随机分为5组,A:正常对照组;B:模型组;C:ABPS高剂量组;D:ABPS中剂量组;E:ABPS低剂量组。8周后,测定空腹血糖(FPG)、空腹胰岛素(FINS)和胰岛素抵抗指数(IR);RT-PCR和western blot检测ADRP基因和蛋白的表达水平。结果显示ABPS治疗组FPG、FINS、IR及ADRP基因和蛋白的表达均低于模型组(P<0.05)。ABPS可抑制ADRP基因表达,降低胰岛素抵抗。 To investigate the effect of Achyranthes bidentata polysaccharides (ABPS) on insulin resistance (IR) and the levels of Adipose differentiation-related protein(ADRP) gene expression in 2-DM rats, Wistar rats were randomly divided into diabetic model group, diabetic model treated with ABPS (100 mg/kg · d) ,ABPS (50 mg/kg · d), ABPS (25 mg/kg · d) group and normal control group. After 8 weeks, the levels of FPG, FINS and IR were measured, and expression of ADRP gene was determined by RT-PCR and western blot. The levels of FPG, FINS, IR as well as the mRNA and protein expression of ADRP in ABPS group were lower than those of diabetic model (P〈0.05). ABPS could decrease IR probably through down regulating the expression of ADRP gene.
