α-干扰素治疗慢性乙型肝炎可促进记忆性CD4^+T细胞群扩增

摘要 目的:探讨α-干扰素对慢性乙型肝炎患者记忆性CD4+T细胞变化的影响及其机制。方法:100例慢性乙型肝炎患者相应的治疗时间采集全血标本,分离外周血单个核细胞,采用FC500流式细胞仪收集和分析外周血记忆性CD4+T细胞亚群。结果:1.在IFN-α治疗过程中,可引起CD4+TCM、CD4+TEM和CD4+TIM细胞比例的规律性变化,即CD4+TCM与CD4+TIM变化规律基本相反,而CD4+TIM与CD4+TEM变化规律基本一致,说明用药时CD4+TCM向CD4+TIM和CD4+TEM转化。2.用药36周时CD4+TCM的增加与CD4+T细胞一致,而停药后CD4+TCM的减少与CD4+T细胞也一致,说明IFN-α导致了CD4+TCM细胞的增殖,从而使CD4+T细胞数量增加。结论:慢性乙型肝炎病毒感染患者用IFN-α治疗时,可引起CD4+TCM、CD4+TEM和CD4+TIM的规律性变化,使得效应CD4+TEM细胞增加;同时,记忆性CD4+T细胞数量增加,其增加原因可能与CD4+TCM细胞数量的增加有关。 Objective:To investigate the influence of IFN-α on memory CD4+T cell subsets of patients with chronic hepatitis B and its mechanism.Methods: Blood samples were collected from 100 patients with CHB prior to treatment(0w) and at time-points,12 w,24 w,36 w,48 w,12 w after treated by IFN-α for 24 w,24 w after treated by IFN-α for 24 w,respectively.PBMC were isolated by density centrifugation using Ficoll-Hypaque.FC500 flow cytometer was used to detect of memory CD4+T lymphocyte subsets.Results: 1.In the course of IFN-α theray,there were regular changes of CD4+TCM,CD4+TEM and CD4+TIM,namely,the change of CD4+TCM and CD4+TIM was reversed while the change of CD4+TIM and CD4+TEM was similar.The results indicated that CD4+TCM could be transformed into CD4+TIM and CD4+TEM.2.At the same time,we found that at the 36th week,following the augment of CD4+TCM,CD4+T cells increased;After withdrawing IFN-α,the percent of CD4+TCM and CD4+T cells decreased.The study indicated IFN-α could result in the augment of percent of CD4+TCM,thereby CD4+T cells increased.Conclusion: In the course of IFN-α theray,there were regular changes of CD4+TCM,CD4+TEM and CD4+TIM,namely,CD4+TCM could be transformed into CD4+TIM and CD4+TEM,so effect memory CD4+T cell increased.At the same time,the percent of memory CD4 +T cells increased,the reason was probably that the augment of CD4+TCM.
