固相萃取-高效液相色谱法测定水中痕量微囊藻毒素 被引量:3

Determination of trace microcystins in water by solid phase extraction-high performance liquid chromatography

摘要 目的:建立固相萃取-高效液相色谱法检测水中微囊藻毒素的方法。方法:水样经固相萃取柱富集,净化,甲醇洗脱,氮吹浓缩,以乙腈-水的体积比为45:55(含0.1%三氟乙酸)为流动相,经C18柱分离,紫外238 nm检测。结果:各组分在0 5.0 mg/L范围内线性良好,0.5μg,1.0μg,4.0μg MC-LR及RR三个水平添加量平均回收率84.3%-87.9%,RSD〈5%。结论:该方法灵敏度高,准确度好,可用于水源水及出厂水中微囊藻毒素的检测。 Objective:To develop a method for determination of microcystin(MC)in water by solid phase extraction-High Performance Liquid Chromatography.Methods:MCs in water were enriched and purified by solid phase extraction,eluated by methanol,the eluate was concentrated by N2 gas.The HPLC separation was carried out with C18 column by UV at 238 nm using acetonitrile-water(include 0.1% TFA) as mobile phase.Results:Tow components showed good linearity in the range of 0-5.0 mg/L,average recovery was 84.3%-87.9%,RSD〈5%.Conclusion:The methods is sensitive,accurate and precise for the determination of MCs in source and effluent waters.
