川芎提取物对大鼠心肌缺血再灌注损伤的影响

Influence of Chuanxiong extract on myocardial ischemia-reperfusion injury in rats

目的观察川芎提取物对大鼠心肌缺血再灌注损伤的影响。方法将40只大鼠随机分为正常组、模型组、川芎小剂量组和川芎组大剂量组各10只,予相应药物灌胃30min后,采用结扎左冠状动脉前降支30 min后再灌注120 min的方法建立大鼠心肌缺血再灌注损伤模型。测定血清丙二醛(MDA)、血清肌酸激酶(CK)、一氧化氮(NO)、一氧化氮合酶(NOS)及超氧化物歧化酶(SOD)含量,处死大鼠取心脏组织经TTC染色测定梗死范围并称重左心室质量,记录心电图以评价川芎提取物对心肌缺血再灌注损伤的影响。结果与模型组比较,川芎提取物组血清CK活性、MDA含量明显降低,NO、NOS、SOD含量增加(P<0.05),心肌缺血面积明显缩小(P均<0.05),心电图中出现ST段异常抬高的导联数(NST)及ST段抬高≥2 mV的总和数(∑ST)和Q波出现的导联数(NQ)均显著减少(P<0.05)。结论川芎提取物对心肌缺血再灌注损伤有保护作用。

Objective It is to observe the influence of Chuanxiong extract（Ligusticum Chuanxiong Hort.） on myocardial ischemia-reperfusion injury in rats.Methods 40 rats were divided into normal group,model group,Chunaxiong low dose group and Chuanxiong high dose group with 10 animals in each group.After treated with homologous drugs 30 min,the model of myocardial ischemia-reperfusion injury was created by left anterior descending coronary artery ligation 30 min and reperfusion 120 min.The levels of serum malondialdehyde（MDA）,serum creatine kinase（CK）,a nitric oxide（NO）,a nitric oxide synthase activity（NOS）,and superoxide dismutase（SOD） were detected,and then,the rats were killed and taken the heart organization to measure the infarct size by TTC staining and weigh the left ventricular.The electrocardiogram was recorded.The influences of Chuanxiong extract on myocardial ischemia-reperfusion injury were evaluated.Results Compared with model group,in Chuanxiong all dose group,the serum levels of CK and MDA were lower（P all0.05）,the levels of NO,NOS and SOD were increased,myocardial ischemic area significantly reduced（P all0.05）.ECG ST segment abnormalities appeared to improve the number of leads（NST） and ST-segment elevation ≥ 2 mV of the total and several（∑ST）,and Q-wave appears the number of leads（NQ） were significantly reduced（P all0.05）.Conclusion Chuanxiong extract has protection on myocardial ischemia-reperfusion injury.
