摘要
目的:测定朝藿定C的平衡溶解度及表观油水分配系数。方法:采用HPLC法测定了朝藿定C在水和7种有机溶剂中的平衡溶解度,及朝藿定C在正辛醇-水/缓冲盐溶液中的表观油水分配系数。结果:28℃时朝藿定C在水中的平衡溶解度为192.66mg·mL-1,表观油水分配系数为1.75(logPapp=0.24),常用有机溶剂甲醇和乙醇对朝藿定C溶解性较好,分别为24.48mg·mL-1和19.84mg·mL-1。结论:朝藿定C水溶性好,在28℃、pH6.8时平衡溶解度达到最大,即295.73mg.mL-1,在正辛醇/pH6磷酸缓冲溶液中油水分配系数最大[3.87(logPapp=0.59)]。因此可以预测朝藿定C有较好的应用价值。
Objective:To determine the equilibrium solubility of epimedin C and its partition coefficients for the n-octanol-water/buffer solution systems.Methods:A high performance liquid chromatography(HPLC) was established and used to detect the concentration of epimedin C in water and seven organic solvents,and also the partition coefficients for the n-octanol-water/buffer solution systems.Results:The equilibrium solubility of epimedin C was 192.66 mg·mL-1 in water at 28 ℃,Papp of epimedin C was 1.75(logPapp=0.24).The higher equilibrium solubility of epimedin C was 24.48 mg·mL-1 and 19.84 mg·mL-1 in methanol and in ethanol.Conclusions:The studies showed that epimedin C was soluble,and when the temperature at 28 ℃,pH 6.8,the equilibrium solubility reached a maximum 295.73 mg·mL-1.It has the highest partition coefficients in n-octanol-water/pH 6 solution systems.Therefore,epimedin C can be predicted has good application value.
