方格星虫粗提物对5/6肾切除大鼠肾小球病变的影响

Effect of Sipunculus nudus preparation on glomerulosclerosis in 5/6 nephrectomized rats

目的观察方格星虫粗提物(SNP)对5/6肾切除大鼠肾小球病变的影响。方法 SD大鼠40只,随机分为假手术组和手术组,手术组行5/6肾切除术并于术后2周随机分为模型组、SNP低剂量组、SNP高剂量组。在给药前及给药4、8、12、16 w末观察大鼠24 h尿蛋白(24 hUPQ)及血肌酐(Scr)的动态变化,普通光镜观察肾小球病理变化,免疫组化法检测肾组织中纤维连接蛋白(FN)、α-平滑肌肌动蛋白(-αSMA)、Wilm肿瘤蛋白-1(WT-1)的表达。结果随着时间的延长,各组大鼠24 hUPQ、Scr均逐渐升高,但同一时间点,治疗组与模型组比较,差异无统计学意义;治疗组大鼠肾小球系膜增生程度、FN及-αSMA表达均较模型组下降;治疗组WT-1表达与模型组比较差异不显著。结论方格星虫粗提物可减轻5/6肾切除大鼠肾小球系膜增生程度、其机制可能与下调FN、-αSMA的表达有关。

Objective To investigate the effects of Sipunculus nudus preparation（SNP） on glomerulosclerosis in 5/6 nephrectomized rats.Methods Forty male SD rats were randomly divided into sham operation group and operation group suffered from 5/6 nephrectomy and further divided into model group,SNP low-dose group,and SNP high-dose group.Before and at the 4,8,12,and 16th weekend after treatment,the levels of 24 hUPQ and Scr were evaluated.The renal histopathology and FN,α-SMA and WT-1 expressions were also detected.Results The 24 h UPQ and Scr of all groups were gradually increased with extension of time,but there was no statistical difference between operation group and treated group at the same time point.Compared with the model group,the glomerular mesangium proliferous degree and the expressions of FN,a-SMA were significantly decreased in treated groups,but no significant difference was found between treated groups and model group in the expression of WT-1.Conclusion SNP can alleviate mesangial ECM deposition and its possible mechanism is to reduce the expressions of α-SMA and FN.
