快速溶剂萃取-液相色谱法测定食品中大豆异黄酮的含量 被引量:2

DETERMINATION OF SOY ISOFLAVONE BY ACCELERATED SOLVENT EXTRACTION AND HIGH PERFORMANCE LIQUID CHROMATOGRAPHY METHOD

摘要 目的建立一种同时测定食品中6种大豆异黄酮[大豆苷元(daidzein)、黄豆黄素(glycitein)、染料木素(genistein)、黄豆黄苷(glycitin)、大豆苷(daidzin)和染料木苷(genistin)]的快速溶剂萃取-高效液相色谱检测方法。方法样品经快速溶剂萃取仪萃取,采用phenomenex LunaC18柱,以0.05%三氟乙酸(TFA)溶液和乙腈为流动相梯度洗脱,二极管阵列检测器检测。结果连续7次测定6种大豆异黄酮,相对标准偏差为1.4%~5.9%。加标回收率82%~112%。在0.01~10μg/ml范围内线性关系良好、最低检出限10μg/kg。结论该方法操作简便,萃取液共存物质对测定方法干扰较小,仪器测定方法准确。适用于大豆异黄酮的检测。 Objective To develop a method based on accelerated solvent extraction and high performance liquid chromatography(HPLC)for determination of 6 soy isoflavones(daidzein,glycitein,genistein,glycitin,daidzin,genistin) in foods.Method Photodiode array detector was employed for detection of these flavoniods by accelerated solvent extraction instrument and Phenomenex Luna? C18 column with 0.05% trifluoroacetic acid(TFA) solution and acetonitrile as mobile phase gradient elution.Results Six kinds of soy isoflavones were determined in samples for 7 times with relative standard deviation 1.4%-5.9%,recovery rate 82%-112%,linear range 0.01-10μg/ml and minimal detection limit 10μg/kg.Conclusion The method is simple,accurate and applicable for determining soy isoflavones in soybean products and the co-existent interference in the extract was relatively small.
