大鼠糖尿病性勃起功能障碍模型的实验研究 被引量:5

Investigation of rat model of diabetic erectile dysfunction

摘要 目的证实链脲佐菌素(STZ)单侧腹腔注射建立大鼠糖尿病性勃起功能障碍模型的可行性。方法 45只SD大鼠随机分为对照组(15只)和实验组(30只)。实验组大鼠腹腔注射STZ 60mg/kg,对照组大鼠腹腔注射相应量的柠檬酸钠一柠檬酸缓冲液,3d后测血糖确定是否成模。此后,每周测血糖1次,8周后用阿朴吗啡(APO)80 μ g/kg颈部皮下注射,观察大鼠阴茎勃起情况。结果实验组糖尿病成模率为93.3%(28/30),在成模后8周的观察期间大鼠的死亡率为7.1%(2/28)。实验组血糖明显高于对照组(P<0.01),体重、阴茎勃起次数、阴茎勃起率则明显低于对照组,两组间差异有统计学意义(P<0.01)。结论 STZ 60mg/kg单侧腹腔内注射能安全有效地建立大鼠糖尿病模型;颈部皮下注射APO 80 μg/kg可有效地筛选出大鼠糖尿病勃起功能障碍。 Objective To study the establishment of rat model of diabetic erectile dysfunction by unilateral intraperitoneal injection with streptozotocin(STZ). Methods All 45 male SD rats were divided into the experimental group ( n=30, unilateral intraperitoneal injection with 60mg/kg STZ) and the control group (n =15, unilateral intraperitoneal injection with sodium citrate buffer). Blood glucose of the treated rats was detected 3 days after injection with STZ or sodium citrate buffer, subsequently, the level of blood glucose was measured in the following each week. Eight weeks later, erectile function of rats was assessed by subcutaneous injection with apomorphine (80 μg/kg). Results 93.3%(28/30) of rats were suffering from diabetes in the experimental group and 7.1% (2/28) were dead after then, Level of blood glucose in experimental group was significantly higher than that in the control group (P〈0.01). But the body weight, the number of penile erection and the rate of erection in the experimental group were significantly lower than those in the control group (P〈0.01). Conclusion Unilateral intraperitoneal injection with STZ (60mg/kg) was an effective method for establishment of diabetic rat model. Subcutaneous injection with Apomorphine (80μg/kg) was an effective method to screen diabetic rat model with erectile dysfunction.
