摘要
为了建立薹菜再生体系进而为转基因工作做准备,以薹菜无菌苗子叶-子叶柄和下胚轴为外植体,研究了适于薹菜再生的外植体类型,以苗龄、AgNO3的质量浓度和植物生长调节剂组合,建立了薹菜的再生体系。结果表明,5 d苗龄的子叶-子叶柄为最佳外植体材料。筛选出最佳培养基为1/2NH4+浓度的MS+BAP5 mg/L+ZT 2 mg/L+NAA 0.7 mg/L+AgNO37.5 mg/L,子叶-子叶柄再生频率达67.8%。
The in vitro cultured hypocotyls and petioles with cotyledon were used as explants to study the effects of explant type,age of seedling,AgNO3 concentration and hormone combination on differentiation frequency of Brassica campestris L.ssp.chinensis Makino var.tai-tsai Hort.The results showed that the petiole with cotyledon explants obtained from 5-day-old seedlings showed the highest shoot-bud differentiation frequency capacity.The best differentiation medium was 1/2NH4+ MS + BAP 5 mg/L + ZT 2 mg / L + NAA 0.7 mg / L + AgNO3 7.5 mg/L.The shoot-bud differentiation frequency was as high as 67.8%.A regeneration system of Brassica campestris L.ssp.chinensis Makino var.tai-tsai Hort was established.
