摘要
目的:研究补阳还五汤对谷氨酸体外诱导脊髓神经元损伤大鼠的保护作用。方法:选用100μmol·L-1的谷氨酸作用于细胞1h,复制谷氨酸诱导原代脊髓神经元调亡的细胞培养模型。实验分为4组,即空白、甲基强的松龙(20μL·mL-1)和补阳还五汤高、低浓度(20、10μL·mL-1)组。药物血清与体外培养5d的原代脊髓神经元共同孵育24h,采用MTT法评价神经元损伤程度。结果:补阳还五汤高、低浓度组神经元线粒体内的琥珀酸脱氢酶的活性明显高于空白组(P<0.01)。结论:补阳还五汤可以部分地抑制谷氨酸的兴奋性神经毒性作用。
OBJECTIVE: To study the protection effect of Buyang huanwu decoction on spinal cord neurons injury induced by glutamate in rats. METHODS: 100 μmol·L^-1 glutamate was used to incubate with primary spinal cord neurons for 1 h, and glutamate-induced primary spinal cord neurons injury model was established. There were four groups, i.e. blank group, methylprednisolone group (20μL·mL^-1), high-concentration Buyang huanwu decoction group (20 μL·mL^-1) and low-concentration Buyang huanwu decoction group (10 μL·mL^-1). Serum containing drugs incubated with primary spinal cord neurons with in vitro 5 days cultivation for 24 h. MTT analysis was used to evaluate the neural injury. RESULTS: The activities of succinic dehydrogenase of mitcohondria in high-concentration and low-concentration Buyang huanwu decoction were significantly higher than in blank group (P〈 0.01). CONCLUSION: Buyang huanwu decoction can partly rescue excitory neurotoxic effects of glutamate.
