大孔吸附树脂结合石硫法提取雷公藤总内酯 被引量:3

Extraction of Total Triptolides from Tripterygium wilfordii by Macroporous Resin Combined with Lime-sulfur Method

摘要 目的:采用石硫法结合大孔吸附树脂提取雷公藤总内酯。方法:以雷公藤内酯醇为对照,采用正交试验选取石硫法提取雷公藤总内酯的工艺条件,再结合大孔吸附树脂对雷公藤总内酯进行富集,并对雷公藤总内酯和雷公藤内酯醇含量进行检测。结果:选取的工艺条件为药材与药液的质量比为1∶3,用石灰乳调pH8后再用10%的硫酸调pH4;结合大孔吸附树脂后,雷公藤总内酯和雷公藤内酯醇的含量分别为85.08、30.8μg·g-1生药。结论:大孔吸附树脂结合石硫法可较好地沉淀杂质,减少大孔吸附树脂用量和有效成分损失,此方法可用于雷公藤叶总内酯的富集。 OBJECTIVE: To extract total triptolides from Tripterygium wilfordii by macroporous resin combined with lime-sulfur method. METHODS: The extraction technology of total triptolides from T. wilfordii was optimized by orthogonal test using triptolide of T. wilfordii as control. The total lactones from T. wilfordii was enriched and cultured by macroporous resin. The content of total triptolides and triptolide of T. wilfordii were determined. RESULTS: Optimized extraction process was as follows: ratio of medicinel materials to solution was 1:3,the pH of mediate was adjusted to 8 using lime milk and then to 4 using 10% sulfuric acid. After using macroporous resin, the content of total triptolides and triptolide of T. wilfordii were 85.08 μg·g^-1 and 30.8μg·g^-1. CONCLUSION: Macroporous resin combined with lime-sulfur method can be used for the precipitation of impurity and reduce the amount of macroporous resin and the loss of active components. The method is applicable in the enrichment of total triptolides of T. wilfordii.
