摘要
目的:建立以高效液相色谱法测定菝葜提取物中槲皮素含量的方法。方法:色谱柱为Kromasil 100-5C18(250mm×4.6mm,5μm),检测波长为370nm,流动相为甲醇-0.4%磷酸(49∶51),柱温为35℃,流速为1.0mL·min-1。结果:槲皮素检测浓度在0.041~0.410μg范围内与峰面积积分值呈良好的线性关系(r=0.9997);平均回收率为97.81%,RSD=1.04%(n=6)。结论:本方法简便、准确,可作为菝葜提取物中槲皮素的含量测定方法。
OBJECTIVE : To establish HPLC method for the content determination of quercetin in extracts from of Smilax china. METHODS: Kromasil 100-5 C18(250 mm×4.6mm,5μm) column was used as chromatographic column and the detection wavelength was set at 370 nm. The mobile phase consisted of methanol-0.4%phosphatic acid (49:51) with flow rate of 1.0 mL·min^-1 and column temperature at 35 ℃.RESULTS: The linear range of quercetin was 0.041μg-0.410 μg(r=0.999 7) with an average recovery of 97.81% (RSD= 1.04%, n= 6). CONCLUSION: The method is simple and accurate for the content determination of quercetin in extracts from S. china.
