改良紫外酶促法在糖尿病患者血浆、尿D-乳酸检测中的应用 被引量:2

摘要 目的:建立一种检测D-乳酸含量的改良紫外酶促法,并探讨其在糖尿病患者血浆、尿D-乳酸检测中的临床应用。方法:对检测血浆、尿D-乳酸含量的商品试剂盒加以改良,并用改良紫外酶促法检测80例糖尿病患者和85例健康成人的血浆和尿液标本中D-乳酸含量。结果:改良紫外酶促法检测血浆、尿D-乳酸在0~200μmol/L的范围内具有良好线性关系;平均批内及批间变异系数分别为3.0%、5.8%,7.1%、6.8%;平均回收率分别为(103.4±8.2)%,(100.1±7.2)%。糖尿病患者血浆D-乳酸的含量[(40.1±24.5)μmol/L]明显高于健康成人[(23.2±10.1)μmol/L],尿中经肌酐校正的D-乳酸含量差异也有显著性[(37.4±5.6)μmol/(L·mMCr)vs(19.3±12.6)μmol/(L·mMCr)]。结论:改良紫外酶促法检测血、尿D-乳酸含量具有较宽的线性范围和可接受的精度及回收率,适合在常规实验室推广应用。糖尿病患者血浆、尿D-乳酸含量明显高于健康成人,检测D-乳酸含量对糖尿病的诊疗有一定的临床意义。
