摘要
采用聚合酶链反应及基因工程技术,克隆卡介苗(BCG)的主要分泌抗原Ag85B基因的5’端第94~211的核苷酸的信号肽序列和构建重组人IL-2穿梭表达质粒,并用酶联免疫吸附法(ELISA)鉴定外源基因在BCG中的表达。结果表明,构建的BCG重组体能表达和分泌人IL-2。此将试用于临床治疗膀胱癌病人。
Polymerase chain reaction and genetic engineering technique were used to clone the signal peptide sequence (position:94 to 211) of antigen 85-B in Mycobacterium bovis BCG and construct the recombinant E.coli-mycobacteria shuttle plasmoid of human interleukin-2(IL-2).Expression of the foreign gene in BCG was detected by enzyme linked immunosorbent assay. The results suggest that the constructed recombinant BCG(rBCG) strain produce and secrete human IL-2 and it will be used for the treatment of superficial bladder cancer.
