摘要
目的:探讨新疆哈萨克族ESCC中miR-203_15甲基化状态与HPV16感染之间的关系及其意义。方法:运用基质辅助激光解析电离时间飞行质谱仪(MALDI-TOFMS)对99例ESCC miR-203_15进行甲基化状态检测;运用半巢式PCR技术对其进行HPV16感染状况检测;运用非参数秩和检验对HPV16(+)与HPV16(-)间miR-203_15甲基化数据进行统计学分析。结果:ESCC HPV16的检出率为32.32%(32/99);HPV16(+)与HPV16(-)间miR-203_15的20个CpG单位的甲基化状态比较,CpG-15.16、CpG_17.18.19.20、CpG_21、CpG_27差异有统计学意义(P<0.05),其中CpG-15.16、CpG_17.18.19.20两个CpG单位平均甲基化率在HPV16(+)组中较高,而CpG_21、CpG_27两个CpG单位平均甲基化率在HPV16(-)组中较高。结论:miR-203_15甲基化与HPV16感染可能共同参与新疆哈萨克族食管鳞状细胞癌的发生,但其内在的关系有待进一步研究。
Objective:Investigate correlation between methylation status of miR-203_15 and HPV16 infection in kazak ESCC of xinjiang province. Methods:The CpG island methylation status of miR-203 in 99 kazak ESCCs was detected by matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS).HPV16 infection status was measured by half-nested PCR.Statistical analysis of the methylation rate between HPV16-positive and HPV16-negative was by nonparametric test.Results:The HPV16-positive rate of 99 ESCCs is 32.32%(32/99).Comparison of the methylation rate between HPV16-positive and HPV16-negative,CpG_15.16,CpG_17.18.19.20,CpG_21,CpG_27 had statistical significance (p0.05),and the mean methylation rates of CpG_15.16,CpG_17.18.19.20 were higher in HPV16-positive and CpG_21,CpG_27 were higher in HPV16-negative.Conclusion:Methylation of miR-203_15 and HPV16 infection may promote the occurrence and progression of ESCC together,but the inherent relationship needs further research.
