子宫平滑肌瘤患者淋巴细胞染色体畸变和脆性位点的研究

RESEARCH ON CHROMOSOME ABERRATION AND FRAGILE SITE IN LYMPHOCYTE OF PATIENTS WITH UTERINE LEIOMYOMA

目的研究子宫平滑肌瘤患者外周血淋巴细胞染色体变化情况。方法分别采用RP-MI1640＋20％小牛血清；Tc199＋20％小牛血清＋Fudr（最终浓度10（－7）mol／L）；TC199＋10％小牛血清＋Aphidicolin（0．2μmol／L）＋乙醇（最终浓度0．02％）培养液进行淋巴细胞培养，对每份样本至少观察100个淋巴细胞中期分裂相的染色体畸变及脆性位点表达的数量、类型及出现频率。结果子宫平滑肌瘤病人的淋巴细胞染色体畸变种类和畸变率明显高于对照组（P＜0．01）。在APC培养条件下，患者组淋巴细胞染色体的脆性位点表达率为70．2％，显著高于对照组的39．8％（P＜0．01）；并且患者组中有20种脆性位点在1／3以上个体中表达，对照组中仅有8种，而且分布特点亦不相同。结论子宫平滑肌瘤患者外周血淋巴细胞染色体不稳定性显著增加。

Objective The chromosome aberration and fragile site in the peripheral blood lymphocyte of 30 patients with uterine leimyoma (UL) were studied. Methods By culture in medium RPM11640＋20% new fetal calf serum; by culture in medium Tcl99 + 10 %new fetal calf serum ＋Fudr (10-7 mol /L ); by culture in medium Tc199 ＋10%new fetal calf serum ＋Aphidicolin (0. 2μmol/L)＋alcohol (0. 02 % ). At least 100 mitosis cells are analysed in each sample to find out the number,type and occurring rate of the chromosome aberration and the development of fragile sites. Results The occurring rate of the chromosome aberration in the patients with UL was significantly higher than that in the controls (P＜0. 01 ). The chromosome aberration types in the patient were more than those in the controls. The rate of fragile site development in the patient(70. 2% ) was significantly higher than that in the controls (39. 8%, P＜0. 01 ). 20 fragile slies developed in more than 1/3 of the patients while only & in the controls. The two groups showed different distribution traits. Conclusion UL patients have marked increase in the chromosome unstability of the peripheral blood lymphocyte.
