Study of significant expression of gp130 molecule on dendritic cells

目的：探讨单核细胞向树突状细胞（ＤＣ）分化成熟时，ｇｐ１３０分子在细胞膜上的表达及其意义。方法：人外周血分离得到的单核细胞在含有ＤＣ分化成熟所需的各种因子的培养液中分别培养４ｄ和７ｄ，然后用流式细胞术测定ＤＣ上ｇｐ１３０分子的表达。结果：ｇｐ１３０分子在单核细胞上低表达，经培养分化成熟成为ＤＣ后，ＤＣ上的ｇｐ１３０分子表达被上调，特别是在ＴＮＦα和抗ＣＤ４０刺激型单克隆抗体存在下。结论：ｇｐ１３０分子表达于ＤＣ前体细胞（单核细胞）并随其分化成熟而增加。

Objective: To investigate expression of gp130 molecule on dendritic cells (DC) when monocytes differentiated and matured into DC. Methods: Monocytes isolated from human peripheral blood were cultured with different combination of cytokines required for DC differentiation and maturation for 4 days and 7 days respectively, the expression of gp130 molecule on DC was analyzed by cytometry. Results: gp130 molecule was low expression on monocytes, and up regulated expression on DC after monocytes were grown and induced into mature DC, especially in the presence of TNFα and CD40 stimulating monoclonal antibody. Conclusion: gp130 molecule is present on monocytes and is increased on mature DC generated from monocytes.
