一种可同时检测抗双链和单链DNA抗体的快速斑点免疫金渗滤试验

A rapid dot immunogold filtration assay for the simultaneous detection of ds DNA and ss DNA antibodies

目的：建立一种可同时检测抗双链ＤＮＡ（ｄｓＤＮＡ）和单链ＤＮＡ（ｓＤＮＡ）抗体的快速斑点免疫金渗滤试验（ＤＩＧＦＡ）。方法：将ｄｓＤＮＡ和ｓＤＮＡ抗原结合在同一检测盒内，采用胶体金标记和快速膜斑点渗滤技术，同时检测抗ｄｓＤＮＡ和ｓＤＮＡ抗体。结果：ＤＩＧＦＡ既可同步又可区别检测两种ＤＮＡ抗体，且检测ｄｓＤＮＡ抗体的敏感性优于酶联免疫吸附法（ＥＬＩＳＡ）。结论：ＤＩＧＦＡ简便、快速、可靠，是一种实用的ＤＮＡ抗体检测方法。

Objective:To develope a rapid dot immunogold filtration assay(DIGFA)for the simultaneous detection of antidouble stranded DNA (ds DNA) and single stranded DNA(ss DNA) antibodies.Methods:ds DNA and ss DNA antigens being incorporated in a sam test kit, a novel DIGFA for the simultaneous detection of anti ds DNA and ss DNA antibodies was established.Results:The test showed that the DIGFA could not only distinguish anti ds DNA from anti ss DNA antibodies but also simplify the procedure of the detection.The DIGFA was more sensitive and specific than enzyme linked immunoabsorbent assay(ELISA).Conclusion:The DIGFA described was rapid,simple and repreducible and was a practical assay for the detection of anti ds DNA and ss DNA antibodies.
